電離輻射誘導腸上皮細胞發生鐵死亡的初步研究

摘要:目的:探究鐵死亡在放射性腸損傷中的作用。方法:從體內體外兩個方面觀察電離輻射能否引起腸道細胞發生鐵死亡。體內實驗:給予人正常腸上皮細胞(HIEC)6Gy X射線照射,分別在照后 0h,24h 和 48h 使用 C11-BODIPY 熒光探針檢測細胞內脂質過氧化水平;利用谷胱甘肽(GSH)試劑盒檢測細胞內 GSH 水平;通過實時熒光定量 PCR檢測前列腺素內過氧化物合成酶2(prostaglandin-endoperoxide synthase 2,PTGS2)mRNA 水平;通過亞鐵離子熒光探針檢測細胞內Fe2+水平;通過透射電子顯微鏡檢測細胞線粒體超微形態。體外實驗:將C57BL/6J小鼠分為對照組和全腹14Gy照射組,照后3.5d 取小腸組織通過HE染色查看小腸組織損傷情況;通過免疫組化檢測谷胱甘肽過氧化物酶 4(Glutathione Peroxidase 4,GPX4)水平;使用電感耦合等離子體質譜(ICP-MS)檢測組織內鐵含量;通過酶標儀檢測組織內MDA和GSH水平。結果:輻照后HIEC細胞內脂質過氧化、PTGS2 mRNA、細胞內亞鐵離子水平升高,GSH水平降低,線粒體嵴缺失或斷裂。小鼠受到14Gy全腹照射后,小腸組織中GPX4、GSH水平降低,PTGS2 mRNA、鐵含量、MDA水平升高。結論:輻射可誘導小腸上皮細胞發生鐵過載和鐵死亡。

Effects of ferroptosis in small intestinal damage induced by ionizing radiation
Abstract:Objective: To identify the impact of ferroptosis in the small intestinal damage induced by ionizing radiation (IR). Methods: We determined the correlation between ferroptosis in small intestine and IR in vivo and in vitro. In vitro experiments: HIEC cells were irradiated by 6 Gy X-rays and collected at 0h, 24h and 48h after irradiation. The level of lipid peroxidation was detected by C11-BODIPY fluorescent staining; The content of glutathione (GSH) was detected by GSH assay kit; The expression of prostaglandin-endoperoxide synthase 2 (PTGS2) mRNA was tested by quantitative real-time PCR (qRT-PCR); Fe2+ fluorescent probe was used to detect the intracellular ferrous ion; The ultrastructure of mitochondria was observed by transmission electron microscope (TEM). In vivo experiment: C57BL/6J mice were randomly divided into two groups: control and irradiation with 14Gy whole abdominal irradiation (WAI). Small intestinal tissues were taken 3.5 days after irradiation. The pathological morphology of intestinal tissues was observed through hematoxylin-eosin (HE) staining, and immunohistochemical staining was used to detect the expression of glutathione peroxidase 4 (GPX4). The iron content in small intestinal tissues was detected by Inductively Coupled Plasma Mass Spectrometry (ICP-MS). The contents of MDA and GSH were measured using a microplate reader. Results: IR boosted the levels of lipid peroxidation, PTGS2 mRNA and Fe2+, downregulated the level of GSH, and induced reduced or absent cristae in HIEC cells. Meanwhile, downregulated expression of GPX4 and GSH together with upregulated the levels of PTGS2 mRNA, MDA and iron were detected in the small intestinal tissues treated with 14 Gy WAI. Conclusion: IR induced iron overload and ferroptosis in intestinal epithelial cells.
