多功能納米脂質體納米材料GOx@Lipid-Cy5.5/GE11的制備及對非小細胞肺癌的診療研究

摘要:目的制備了一種熒光靶向載藥脂質體納米材料GOx@Lipid-Cy5.5/GE11,并在非小細胞肺癌細胞中研究其熒光示蹤和體外抗腫瘤的能力。方法通過薄膜分散法和正交試驗探索脂質體最佳合成工藝,并利用點擊化學制備并表征了GOx@Lipid-Cy5.5/GE11。利用細胞攝取和細胞毒性實驗在非小細胞肺癌A549細胞中驗證了GOx@Lipid-Cy5.5/GE11的親細胞特性和抗腫瘤作用。結果優化工藝并成功制備出了一種多功能靶向載藥脂質體納米材料GOx@Lipid-Cy5.5/GE11,其水合粒徑為100~200 nm,分散性良好,包封率>90%。細胞實驗表明,其能夠被細胞高效攝取并對非小細胞肺癌細胞具有明顯毒性。結論 GOx@Lipid-Cy5.5/GE11是一種多功能脂質體復合納米材料,在 NSCLC診療中具有廣闊的應用前景。

Multifunctional nanoliposome nanomaterials GOx@Lipid-Preparation of Cy5.5/GE11 and its diagnosis and treatment of non-small cell lung cancer
Abstract:Objective: prepare a fluorescent targeting liposome nanomaterial GOx@Lipid-Cy5.5/GE11, and its fluorescence tracing and antitumor ability in vitro were studied in non-small cell lung cancer cells. Methods: the synthetic process of liposomes was explored through thin film dispersion method and orthogonal test, and the liposomes were prepared and characterized by click chemistry GOx@Lipid-Cy5.5/GE11. The cytotoxic properties and antitumor effects of GOx@Lipid-Cy5.5/GE11 was verified in non-small cell lung cancer A549 cells by cell uptake and cytotoxicity experiments. Results: a multifunctional targeting liposome nanomaterial was successfully prepared by optimizing the process GOx@Lipid-Cy5.5/GE11, the hydrated particle size is 100~200 nm, the dispersion is excellent, and the entrapment efficiency is > 90%. Cell experiments show that it can be efficiently ingested by cells and has obvious toxicity to non-small cell lung cancer cells. Conclusion: GOx@Lipid-Cy5.5/GE11 is a multifunctional liposome composite nanomaterial, which has broad application prospects in the diagnosis and treatment of NSCLC.
